Descripción : (Nitric oxide and reactive nitrogen species: Basic concepts and biologic importance) Nitric oxide (NO) is a free radical synthesized from the amino acid L-arginine by a family of enzymes, the nitric oxide synthases (NOS). There are three isoforms of NOS named neuronal NOS (nNOS or NOS type I), endothelial NOS (eNOS or NOS type III) and inducible NOS (iNOS or NOS type II). nNOS and eNOS are constitutively expressed in mammalian cells which synthesize NO⋅ in response to the increase in intracellular calcium level. iNOS activity is independent of the calcium levels in the cell. Many biological functions of NO⋅ depend on activation of soluble guanylate cyclase, resulting in elevated production of cyclic guanosin monophosphate (cGMP) levels and activation of cGMP dependent protein kinases which induce diverse biological responses such as smooth muscle relaxation and vasodilation, inhibition of platelet aggrega tion and neurotransmission. NO is able to react with thiol groups (S-nitrosylation) forming S-nitrosothiols; however NO reactivity is low despite is a free radical. NO is the precursor of reactive nitrogen species (RNS) such as peroxynitrite (ONOO–), peroxynitrous acid (ONOOH), nitrogen dioxide radical (NO2 ), dinitrogen trioxide (N2O3) and nitryl chloride (NO2Cl). NO overproduction is associated with enhanced production of RNS, which induce structural damage to multiple biomolecules including proteins and deoxyribonucleic acid. The presence of 3-nitrotyrosine is considered a footprint of RNS.
